A biopsy is the removal of tissue or cells from a living patient for microscopic, immunohistochemical, or molecular examination, providing the definitive basis for diagnosing and classifying disease. Techniques range from fine-needle aspiration cytology and core-needle sampling to incisional, excisional, and bone marrow biopsy, each selected according to the lesion, anatomic site, and diagnostic question. In hematology and oncology, biopsy underpins the diagnosis of malignancy by enabling histopathologic confirmation, tumor grading and subtyping, assessment of margins, and increasingly the detection of biomarkers and molecular alterations that direct prognosis and targeted therapy. Cytologic evaluation of lymphoid lesions, for example, must resolve atypical or suspicious categories, while immunohistochemical and chemical analyses refine characterization of solid tumors and bone marrow disease.
